The study investigates the vulnerability of layer 5a corticostriatal neurons in Huntington's disease (HD) through the innovative technique of serial fluorescence-activated nuclear sorting (sFANS). While layer 5a neurons are particularly susceptible, other deep layer neurons demonstrate greater resilience despite shared mHTT CAG repeat expansion. The findings suggest that altered synaptic function and disrupted corticostriatal connectivity may play a crucial role in the selective vulnerability of layer 5a neurons.
